Details TRH Receptors: Is the Consensus View on CNS Receptors Correct?

SUMMARY: The central nervous system (CNS) effects in rodents of thyrotropin-releasing hormone (TRH) have been thought to be mediated by TRH receptor type 2 (TRH-R2) whereas the endocrine effects by TRH-R1. I will present evidence using TRH-R1 knockout (R1ko), R2ko and R1/R2ko mice that the primary mediator of all known TRH effects is TRH-R1, which is highly homologous to the single human TRH receptor. These findings suggest that a small, lipophilic, drug-like agonist for TRH receptor may have therapeutic effects in several CNS disorders. I will then describe an approach to the development of drug-like agonists for the human TRH receptor.
